Calcitonin-salmonCalcitonin analogue
Calcitonin-salmon reduces the activity of bone-resorbing cells and can lower blood calcium. Its history in osteoporosis is different from its use in selected calcium and bone disorders.
What is Calcitonin-salmon?
Salmon calcitonin is a medicinal form of calcitonin based on the salmon hormone. Injectable and nasal preparations have different histories and uses.
How it works
Calcitonin-receptor activity inhibits bone-resorbing osteoclasts and can lower blood calcium. Its effects therefore depend on the condition being treated.
Effects studied
Bone and calcium disorders
Calcitonin has been studied in osteoporosis and used in selected calcium and bone disorders. An older osteoporosis study assessed fractures. 1
Evidence: Evidence and indications are formulation-specific; the older trial does not establish current availability or preferred use.
What does the evidence show?
Older osteoporosis trials and regulatory safety-review lead
Why a salmon hormone appears in medicine
The medicine is based on the salmon form of calcitonin, usually produced synthetically. It acts at calcitonin receptors and suppresses osteoclast activity: the cells that break down bone. A fall in bone resorption or blood calcium is a pharmacological effect, not automatically evidence of improved long-term bone health.
Several uses should not be merged
Injectable and nasal preparations have been studied or used in different settings, including high blood calcium, Paget’s disease and osteoporosis. They do not share an interchangeable benefit-risk profile. A study of nasal treatment for fractures cannot answer the same question as short-term treatment of a calcium emergency.
What changed the osteoporosis discussion
An older fracture trial remains part of the evidence history. Later regulatory reviews examined a possible malignancy signal with longer-term exposure. A positive result for one outcome has to be considered alongside that safety concern. The current local product information is essential before describing any preparation as an available osteoporosis option. 1 2
The useful comparison
Calcitonin affects bone. The clinical question is whether, over the intended treatment period, it improves the relevant outcome compared with other available treatments. Nausea, flushing and hypersensitivity are additional clinical considerations, especially when treatment is given by injection.
Safety & status
Nausea, flushing and hypersensitivity are relevant adverse effects. A European regulatory review raised concerns about malignancy with long-term exposure. Current product restrictions need a country-level check. 2
Country and product status
Product indications and availability vary by country; current EU and Swiss status has not been independently checked for this draft.
Frequently asked questions
Are nasal and injected calcitonin interchangeable?
No. Route, indication and regulatory restrictions need to be considered for the specific product.
